Best way to travel in Ecuador

Other than flying is it possible to go from Guayaquil to Quito on a bus and on a good highway? Is it on narrow roads hanging off a cliff? Is it safe or are there many bus robberies on the higway as the state dept suggests.
How driving time is there from Quito to Cuenca? Is it on a good road? Are there public buses? Can you board them with a small suitcase?
