Getting from Quito to Cuenca

Hi everyone,
I'd like to go to Cuenca in January. I'm going to fly from Frankfurt (Germany) to Quito. Now I don't know how I should get from Quito to Cuenca?! What can you recommend me? Should I take a plane, train or bus?
